The arithmetic

Concentration = mass ÷ volume
10 mg into 2 mL → 5 mg/mL, i.e. 5000 mcg/mL.
Volume = target mass ÷ concentration
250 mcg at 5000 mcg/mL → 0.05 mL.
1 mg = 1000 mcg
Vials are labelled in mg; aliquots are usually discussed in mcg.
Diluent volume is what you added
Lyophilized powder contributes negligible volume at these masses.
